The body made white blood cells called lymphocytes to fight infection or foreign substances which have entered the body. Following an organ transplant your body's immune system recognises the new organ as 'foreign' to you and will try to attack it with lymphocytes. Pograf contain Tacrolimus works by stopping the body from producing these lymphocytes and prevents the attack on the transplanted organ.
Tacrolimus is also available as an ointment which is used in the treatment of eczema. There is a separate information leaflet available about this called Tacrolimus (topical).
Prograf, contain Tacrolimus should be used with caution in :
- Liver problems
- Diarrhoea for more than one day
- Patients requiring vaccinations
- Patients having exposure to the sun and sunbeds
Tacrolimus ointment should not be used in :
- Hypersensitivity to tacrolimus, sirolimus, any of the other ingredients in the capsule granules or ointment, or any macrolide antibiotic.
- Children under the age of 2 years
- Contaminated lesions
- Previous or current skin cancers
- Problems with your immune system
- Inherited skin disease, such as Netherton's syndrome
- People who suffer from red and scaly skin all over your body (erythroderma)
- Light therapy
- Pregnanacy
Dosage :
- Before starting this treatment, read the manufacturer's printed information leaflet from inside the pack.
- Take this tablet exactly as your doctor has told you.
- Take this tablet when your stomach is empty. This means an hour before food or 2-3 hours after food.
- It is important that you do not change the brand of tacrolimus that you are taking except on the advice of your transplant specialist. Different brands release different amounts of tacrolimus and if you swap formulations this can either stop the medicine from working properly or lead to side-effects.
- Try to take at the same times each day to avoid missing any doses. If you forget to take a dose, take it as soon as you remember unless it is nearly time for your next dose, in which case leave out the missed dose. Do not take two doses together to make up for a forgotten dose.
- Do not drink grapefruit juice while you are taking tacrolimus as it can increase the levels of tacrolimus in your body.
- Do not stop taking it unless your doctor tells you to do so. Stopping taking it will increase the risk of your transplanted organ being rejected.
Storage :
Store tacrolimus ointment below 25°C.
Store tacrolimus tablets or suspension below 25°C in the original package to protect from light and moisture.
